Because my employer say me that i need to under go with medical checkup from Authorize hospitals list given by us consulate?
No need for H1B stamping.
No, that is not true for H1B stamping. Ask them to show the requirements. Check USTraveldocs, it will tell clearly the requirements for stamping.

Do i require canceled Passport during interview?
as I hare passport renewed but I don’t have previous canceled passport.
if Yes then what process I have to do? as I don’t have my previous passport.
This is my first time stamping.

Not really required as it is not valid anymore. They usually will not ask, unless it has any visas in it. So, not to worry. Carry a copy of the previous passport cancelled, if you have it to be safe.
