My H4 is expiring in May 2021. My spouse’s H1b is expiring in August 2021.
Should I file my H4&EAD(I am eligible) right now in regular processing or should I wait until to file with his H1b extension in premium processing?

You can file it now or later. The only issue with filing it now is that you will only get your H4 and EAD until Aug 2021. The best thing to do it wait for few more months and then apply for extension as well as other things together…Otherwise, for the sake of two months, you again need to reapply.
